**dEGF4KDEL** is an investigational, single-chain deimmunized bispecific ligand-directed toxin developed at the University of Minnesota. It fuses human epidermal growth factor and interleukin-4 targeting ligands to a truncated, deimmunized *Pseudomonas aeruginosa* exotoxin A payload with a C-terminal KDEL retention sequence. The molecule binds epidermal growth factor receptor and interleukin-4 receptor-positive tumor cells, is internalized, and delivers the exotoxin catalytic domain to inhibit protein synthesis and induce cell death. It showed preclinical antitumor activity in pancreatic cancer xenograft models, including inhibition of orthotopic MIA PaCa-2 tumors and metastasis, and related publications describe activity in breast cancer and malignant mesothelioma models.
