Overview

Dehydroevodiamine is a naturally occurring quinazoline alkaloid isolated from *Evodiae Fructus* (the fruit of Evodia rutaecarpa or related species), a well-known herb in traditional Chinese medicine. It exhibits diverse pharmacological activities, including anti-inflammatory, antiarrhythmic, anti-amnesic, neuroprotective, hypotensive, antitumor, and anti-depressant effects[6][10][13][15][16][1]. Mechanistically, dehydroevodiamine acts as an inhibitor of acetylcholinesterase and butyrylcholinesterase, providing a potential benefit for cognitive impairment, particularly in Alzheimer’s disease and stress-induced memory deficits[1][7][4][12]. It also inhibits aldehyde dehydrogenase 3A1, has anti-proliferative effects in liver cancer by reducing glucose metabolism and glycolysis, and demonstrates antiarrhythmic properties, partly due to inhibition of hERG (human Ether-à-go-go-Related Gene) potassium channels and suppression of inflammatory pathways such as NF-κB[1][11][17]. Dehydroevodiamine improves memory, enhances neuronal plasticity, and reduces tau hyperphosphorylation[4][12]. It is studied as a central nervous system agent and is also under investigation for chronic gastritis, gastric ulcers, and rheumatoid arthritis[6]. Major toxicological considerations include possible pro-arrhythmic effects and impairment of bile acid homeostasis without significant hepatotoxicity[6]. Dehydroevodiamine has not been approved as a pharmaceutical drug and remains primarily a research compound.
