Overview

This is a combination regimen of four investigational and approved drugs for tuberculosis (TB): delamanid, bedaquiline, OPC-167832 (quabodepistat), and sutezolid. Delamanid is a nitroimidazole that inhibits mycolic acid synthesis in mycobacterial cell walls. Bedaquiline is a diarylquinoline that targets ATP synthase, disrupting energy production in Mycobacterium tuberculosis. OPC-167832 (quabodepistat) is a novel carbostyril derivative that inhibits decaprenylphosphoryl-beta-D-ribose 2'-oxidase (DprE1), an essential enzyme for cell wall biosynthesis in M. tuberculosis[1][2][3]. Sutezolid is an oxazolidinone antibiotic related to linezolid, which inhibits protein synthesis by binding to the bacterial 50S ribosomal subunit. This combination aims to provide potent bactericidal activity against drug-sensitive and multidrug-resistant TB strains with the potential to shorten treatment duration[1][2][3].
