Delavirdine + nucleoside reverse transcriptase inhibitors is a combination antiretroviral therapy comprised of the non-nucleoside reverse transcriptase inhibitor (NNRTI) delavirdine and one or more nucleoside reverse transcriptase inhibitors (NRTIs; such as zidovudine, lamivudine, didanosine, or zalcitabine). Delavirdine works by binding allosterically and non-competitively to HIV-1 reverse transcriptase, inhibiting its RNA-dependent and DNA-dependent DNA polymerase activities, thereby blocking viral replication. NRTIs act as chain terminators when incorporated into viral DNA by reverse transcriptase, further inhibiting HIV replication. This combination provides synergistic antiretroviral effects and is indicated for the treatment of HIV-1 infection, frequently as part of triple therapy regimens, especially in cases requiring first-line antiretroviral therapy or salvage treatment after the failure of other regimens[1][2][3][4][5][6][7].
