**delNS1-GM-CSF** is an investigational, replication-competent oncolytic influenza A virus based on the A/Puerto Rico/8/1934 influenza A backbone. It contains a partial deletion in the nonstructural protein 1 gene and an inserted granulocyte-macrophage colony-stimulating factor coding sequence. The construct is intended to preferentially replicate in and lyse hepatocellular carcinoma cells while producing GM-CSF locally to enhance antitumor immune activation. In preclinical hepatocellular carcinoma models, intratumoral administration inhibited tumor growth, increased tumor-associated CD4-positive and CD8-positive T-cell responses, and showed enhanced activity when combined with programmed cell death protein 1 blockade. Reported mechanistic analyses associated its antitumor activity with modulation of the Janus kinase 2-signal transducer and activator of transcription 3 pathway.
