Delpacibart zotadirsen (formerly AOC 1044) is an antibody-oligonucleotide conjugate (AOC) developed by Avidity Biosciences for the treatment of Duchenne muscular dystrophy (DMD) in patients with mutations amenable to exon 44 skipping. The drug consists of a monoclonal antibody that binds to the transferrin receptor 1 (TfR1) conjugated to a phosphorodiamidate morpholino oligomer (PMO) antisense oligonucleotide. By targeting TfR1, the AOC facilitates the delivery of the PMO into skeletal muscle and heart tissue. Once inside the cells, the PMO induces skipping of exon 44 in the dystrophin pre-mRNA, restoring the reading frame and allowing for the production of a truncated but functional dystrophin protein. It is currently in Phase 1/2 clinical development (EXPLORE44 trial).
