Delta-24-GREAT (Glucocorticoid Receptor Enhanced Activity of T cells) is an armed, replication-competent oncolytic adenovirus designed for the treatment of malignant brain tumors, such as glioblastoma, and other solid tumors like metastatic breast cancer. It is a genetically modified derivative of Delta-24-RGD (DNX-2401) that has been engineered to express the immune co-stimulatory ligand GITRL (Glucocorticoid-induced TNFR-related protein ligand). The viral backbone contains a 24-base pair deletion in the E1A gene, restricting its replication to retinoblastoma (Rb) pathway-deficient cancer cells, and incorporates an RGD-4C motif in the fiber HI-loop to enhance infectivity via integrin binding. Ectopic expression of GITRL on the surface of infected tumor cells stimulates tumor-infiltrating lymphocytes, promoting T-cell recruitment, activation, and the generation of antigen-restricted anti-tumor memory.
