Delta-24-RGD-IL15 is an experimental second-generation oncolytic adenovirus (OA) engineered for the treatment of solid tumors, including gliomas and melanomas. It is based on the Delta-24-RGD platform, which features a 24-base-pair deletion in the E1A region to ensure selective replication in cancer cells with a deficient retinoblastoma (Rb) pathway and an RGD (Arg-Gly-Asp) motif in the fiber knob to enhance viral entry via integrins. This specific iteration is further modified to express Interleukin-15 (IL-15), a cytokine that stimulates the activation and proliferation of T cells and natural killer (NK) cells and promotes the persistence of CD8+ memory T cells. By combining direct viral oncolysis with IL-15-mediated immune stimulation, Delta-24-RGD-IL15 aims to enhance anti-tumor immunity within the tumor microenvironment.
