Delta-24-RGDOX-IL15 is a next-generation oncolytic adenovirus that co-expresses two immune-stimulating molecules: OX40 ligand (OX40L) and interleukin-15 (IL-15). It is built upon the Delta-24-RGD platform, which is a conditionally replicating, tumor-selective oncolytic adenovirus with enhanced infectivity. The virus is designed to selectively replicate in and kill cancer cells while simultaneously activating the immune system against tumors. The addition of IL-15 to the virus is strategically important as IL-15 activates T and NK cells and promotes the persistence of CD8+ memory T cells, potentially enhancing long-term anti-tumor immunity. In preclinical studies, Delta-24-RGDOX-IL15 has shown superior efficacy compared to earlier generations of oncolytic adenoviruses in inhibiting tumor growth in both treated tumors and untreated distant tumors, suggesting its potential to generate systemic anti-tumor immunity that can target metastatic disease.
