DeltaRex-G is a tumor-targeted gene therapy based on an amphotropic retrovector derived from murine leukemia virus (MLV). It encodes a cytocidal CCNG1 inhibitor gene that blocks the expression of cyclin G1 (CCNG1), thereby arresting the cancer cell cycle in the G0–G1 phase and inducing apoptosis in rapidly dividing cells. The vector is engineered to display a collagen-matrix-binding decapeptide on its gp70 envelope protein, enabling it to selectively bind to abnormal collagenous proteins within the tumor microenvironment. This targeting allows for delivery of its payload specifically into cancer cells, proliferating tumor-associated fibroblasts, and neoangiogenic endothelial cells. DeltaRex-G has been studied primarily for advanced cancers including pancreatic adenocarcinoma, breast cancer (including triple-negative and HR+Her2+ subtypes), sarcoma, squamous cell carcinoma of skin, and B-cell lymphoma. It has also received emergency use authorization for COVID-19-induced acute respiratory distress syndrome due to its ability to inhibit hyperactive immune cells[1][2][3][4][5].
