Deltex1 protein lipid nanoparticles (Dtx1-LNPs) are a preclinical-stage therapeutic designed to treat T-cell acute lymphoblastic leukemia (T-ALL). The drug consists of the wildtype, full-length Deltex1 (Dtx1) protein, an E3 ubiquitin ligase, encapsulated within targeted lipid nanoparticles for delivery to human T-ALL cells. Deltex1 acts as a natural inhibitor of Notch1 signaling by interacting with the Transcriptional Activation Domain (TAD) of the Notch1 receptor and promoting its degradation. This mechanism addresses the hyperactive Notch1 signaling that drives leukemia in the majority of T-ALL patients. By utilizing a targeted LNP delivery system, Dtx1-LNPs aim to overcome the systemic toxicity and lack of tissue specificity associated with existing Notch1-targeting therapies like γ-secretase inhibitors and anti-Notch1 antibodies.
