DEM301 is a novel antibody-drug conjugate (ADC) designed to target human Cadherin-17 (CDH17), a cell-surface glycoprotein highly expressed in gastrointestinal (GI) cancers, including colorectal, gastric, and pancreatic tumors. The agent consists of a humanized, afucosylated monoclonal antibody conjugated to a potent topoisomerase I inhibitor payload via a cleavable linker. By targeting CDH17, DEM301 facilitates the selective delivery of the cytotoxic payload to tumor cells, where it induces DNA damage and cell death. Preclinical studies have demonstrated that DEM301 exhibits robust anti-tumor activity, including complete and partial responses in various xenograft models, while maintaining a favorable safety profile in humanized transgenic mouse models. The afucosylated nature of the antibody also enhances its ability to mediate antibody-dependent cellular phagocytosis (ADCP), further contributing to its anti-tumor efficacy.
