demcizumab

Demcizumab is a humanized IgG2 monoclonal antibody that targets Delta-like ligand 4 (DLL4), a key activator of the Notch signaling pathway. By binding to DLL4 and preventing its interaction with Notch-1 and Notch-4 receptors, demcizumab inhibits Notch-mediated signaling and gene transcription. This mechanism impedes tumor angiogenesis and may block the growth of cancer stem cells as well as prevent new blood vessel formation required for tumor growth. Demcizumab was developed by OncoMed Pharmaceuticals in collaboration with Celgene for use in solid tumors such as pancreatic cancer, non-small cell lung cancer (NSCLC), ovarian cancer, colorectal cancer, fallopian tube cancer, peritoneal cancer, and other solid tumors. It has received orphan drug designation for pancreatic cancer but development has been discontinued across all indications.
