Drug intelligence / Profile preview
This combination consists of **demcizumab** (a humanized monoclonal antibody targeting Delta-like ligand 4 or DLL4 in the Notch signaling pathway), **gemcitabine** (a nucleoside analog cytotoxic agent inhibiting DNA synthesis), and **nab-paclitaxel** (nanoparticle albumin-bound paclitaxel, a microtubule-stabilizing agent). **Demcizumab** blocks DLL4, inhibiting Notch signaling involved in cancer stem cell maintenance and angiogenesis, leading to decreased cancer stem cell frequency and tumor growth. **Gemcitabine** interferes with DNA replication, leading to cancer cell death. **Nab-paclitaxel** disrupts microtubule dynamics and increases intratumoral gemcitabine by reducing cytidine deaminase (CDA) via reactive oxygen species, thereby synergizing with gemcitabine for enhanced cytotoxicity. The combination is studied as a first-line therapy in metastatic pancreatic cancer and being evaluated in clinical trials for efficacy and safety[1][2][3][4][7][10].
