Overview

**Demcizumab + pembrolizumab** is an experimental combination therapy of two monoclonal antibodies for cancer treatment. Demcizumab is a humanized monoclonal antibody targeting Delta-like ligand 4 (DLL4), a component of the Notch signaling pathway, and is believed to inhibit tumor angiogenesis and modulate immune response by decreasing monocytic myeloid-derived suppressor cells (MDSCs) in tumors. Pembrolizumab is a humanized IgG4 monoclonal antibody (brand name Keytruda) that blocks the programmed cell death protein 1 (PD-1) on lymphocytes, thereby inhibiting an immune checkpoint and enhancing anticancer immune activity. This combination aims for a double blockade of cancer-induced immune suppression. It has been evaluated in a Phase 1b clinical trial in advanced/metastatic solid tumors, including specific cohorts for non-small cell lung cancer (NSCLC), anti-PD1 refractory cancers, and castrate-resistant prostate cancer[1][2].
