Demplatin pegraglumer is a micellar drug formulation composed of the platinum-based chemotherapeutic cisplatin incorporated into polymeric micellar nanocarriers using an mPEG-poly(l-Glu) diblock copolymer with a 1,3-propanediyl linkage. This design aims to improve the release and delivery of cisplatin, a standard treatment for many cancer types, by enhancing its pharmacokinetics and reducing toxicity. The mechanism of action is based on the activity of cisplatin, which forms DNA crosslinks that inhibit DNA replication and transcription, leading to cell cycle arrest and apoptosis in rapidly dividing tumor cells. Demplatin pegraglumer has been investigated primarily for neoplasms such as pancreatic cancer and head and neck cancers. It has completed Phase 3 trials in pancreatic neoplasms and Phase 2 trials in other solid tumors[1][2][4][7].
