DEN1-80E is a recombinant subunit vaccine antigen consisting of a truncated envelope (E) glycoprotein from dengue virus serotype 1. It is designed as part of monovalent or tetravalent dengue vaccine formulations, where similar proteins from all four dengue serotypes are combined. The protein is produced recombinantly (commonly in Drosophila S2 cells), and formulated with adjuvants such as Alhydrogel or ISCOMATRIX to enhance immunogenicity. In preclinical and early clinical studies, DEN1-80E has been shown to induce neutralizing antibodies against DENV-1 and provide protection against viremia in animal models. Clinical trials have demonstrated that the vaccine induces modest but detectable neutralizing antibody responses in flavivirus-naïve adults, with good tolerability[5][6][1].
