Drug intelligence / Profile preview
Dendrimer-TTM020 (D-TTM020) is an investigational nanoparticle polymer-drug conjugate consisting of the glutamine antagonist and 6-diazo-5-oxo-L-norleucine (DON) prodrug, TTM020, covalently linked to a hydroxyl-terminated poly(amidoamine) (PAMAM) dendrimer. Co-developed by researchers at Johns Hopkins University and the Institute of Organic Chemistry and Biochemistry of the Czech Academy of Sciences (IOCB Prague), Dendrimer-TTM020 is designed to selectively target activated microglia in the central nervous system. Upon systemic administration, the dendrimer carrier crosses the blood-brain barrier and is selectively engulfed by inflamed microglia, where it releases the active glutamine antagonist to inhibit glutaminase (GLS1) activity. This localized mechanism of action mitigates the severe gastrointestinal and systemic toxicities typically associated with free glutamine antagonists. Dendrimer-TTM020 is currently in preclinical development for the treatment of neurodevelopmental and neuropsychiatric disorders, including Rett syndrome and chronic stress-induced depression.
