This autologous dendritic cell vaccine, developed by the H. Lee Moffitt Cancer Center and Research Institute, is a cell-based immunotherapy designed for the neoadjuvant treatment of high-risk soft tissue sarcoma. The therapeutic process involves harvesting a patient's own dendritic cells (DCs) via apheresis, followed by ex-vivo expansion and culture under standardized GMP conditions. The treatment protocol consists of four intratumoral injections of approximately 10 million DCs each, administered in conjunction with neoadjuvant external beam radiation therapy (EBRT) and followed by surgical resection. The mechanism of action leverages radiation-induced immunogenic cell death to release tumor-associated antigens within the tumor microenvironment; the injected DCs then capture these antigens and migrate to regional lymph nodes to prime and activate tumor-specific T-lymphocytes. This approach aims to overcome the typically low immunogenicity of sarcomas and establish a durable anti-tumor immune response.
