Overview

The dendritic cell survivin vaccine (specifically DC:AdmS) is an autologous cancer vaccine composed of a patient's own dendritic cells that have been genetically modified using a replication-deficient adenoviral vector to express the survivin protein. Survivin (BIRC5) is a member of the inhibitor of apoptosis protein (IAP) family that is highly expressed in various malignancies, including multiple myeloma and glioblastoma, while being nearly absent in normal adult tissues. By presenting survivin-derived peptides via MHC class I and II molecules, the vaccine is designed to prime and expand survivin-specific CD8+ and CD4+ T cells. In clinical settings, particularly for multiple myeloma, it is often administered in conjunction with autologous hematopoietic cell transplantation (HCT) to leverage the period of lymphopenia-induced homeostatic proliferation to enhance the anti-tumor immune response.
