Overview

This investigational cellular immunotherapy, developed at Beth Israel Deaconess Medical Center, consists of a vaccine created by the fusion of autologous dendritic cells (DCs) with patient-derived renal cell carcinoma (RCC) cells. The fusion process, typically achieved via polyethylene glycol or electrofusion, combines the potent antigen-presenting capabilities of DCs with the broad array of tumor-associated antigens present in the patient's own cancer cells. This hybrid cell is designed to present these antigens in the context of DC-mediated costimulatory molecules, thereby inducing a robust, polyclonal anti-tumor T-cell response. The vaccine is administered subcutaneously in combination with granulocyte-macrophage colony-stimulating factor (GM-CSF), which serves as an adjuvant to recruit and activate endogenous antigen-presenting cells at the injection site, further enhancing the immune response. Clinical evaluation in Phase I/II trials has demonstrated that the approach is well-tolerated and capable of inducing immunologic and clinical responses, including partial responses and disease stabilization, in patients with metastatic RCC.
