Overview

Dendritic cell vaccination + temozolomide is a combination therapy used primarily in the treatment of glioblastoma. The regimen combines autologous dendritic cell vaccines—often loaded with tumor lysate or specific tumor antigens—with the alkylating chemotherapy agent temozolomide. Dendritic cells are harvested from the patient, pulsed ex vivo with tumor antigens (such as autologous tumor lysate or mRNA), and then re-administered to stimulate an anti-tumor immune response by activating cytotoxic T lymphocytes against glioblastoma cells[5][7][9]. Temozolomide is a standard-of-care oral chemotherapy for glioblastoma that induces DNA damage in rapidly dividing tumor cells. The rationale for combining these therapies is to enhance anti-tumor immunity while maintaining cytotoxic pressure on the cancer[1][4]. Clinical trials have shown that this combination can be safe and well tolerated, with some studies reporting improved progression-free and overall survival compared to standard therapy alone[2][5][9].
