Overview

The dendritic cell vaccine developed at Karolinska University Hospital is an investigational autologous cellular immunotherapy designed for the treatment of metastatic melanoma. This therapeutic approach involves the ex vivo maturation of a patient's own dendritic cells, which are then used to vaccinate the patient to prime and expand tumor-specific T-cell responses. In clinical settings, notably in the Phase I trial NCT01946373, the vaccine has been evaluated as part of a multi-modal regimen that includes adoptive cell transfer of tumor-infiltrating lymphocytes (TILs). This regimen typically follows lymphodepleting chemotherapy with cyclophosphamide and fludarabine and is supported by the administration of interleukin-2 (IL-2) to enhance the persistence and anti-tumor activity of the transferred and vaccine-stimulated T cells.
