Dendritic cell vaccine (Mackay Memorial Hospital) is an autologous, cell-based cancer immunotherapy developed for the treatment of hepatocellular carcinoma (HCC). The vaccine is produced by isolating monocytes from the patient's peripheral blood, differentiating them into dendritic cells, and pulsing them with autologous tumor lysates derived from the patient's own tumor. These dendritic cells are matured using a cytokine cocktail containing recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF) and interleukin-4 (rhIL-4). The matured, antigen-loaded dendritic cells are then reintroduced into the patient to stimulate a specific cytotoxic T lymphocyte (CTL) response against HCC cells. Research at Mackay Memorial Hospital has also investigated the use of low-dose radiation to enhance the immunogenicity of the tumor cells used for the lysate.
