This is a combination immunotherapy consisting of a dendritic cell vaccine and denileukin difitox. Dendritic cell vaccines are autologous or allogeneic antigen-presenting cells loaded with tumor-associated antigens, designed to stimulate an anti-tumor immune response by activating T cells against cancer-specific targets[1][2][5]. Denileukin difitox is a recombinant fusion protein composed of interleukin-2 (IL-2) fused to diphtheria toxin, which selectively targets and depletes CD25-expressing regulatory T cells (Tregs), thereby reducing immune suppression in the tumor microenvironment[8][9]. The rationale for combining these therapies is that depletion of Tregs may enhance the efficacy of dendritic cell vaccination by allowing more robust cytotoxic T lymphocyte responses against tumors[5][7]. This combination has been investigated in clinical trials for advanced cancers such as melanoma and renal cell carcinoma, with mixed results regarding enhancement of vaccine-induced immunity; some studies report increased tumor-specific cytotoxic T lymphocytes, while others show limited benefit depending on dosing and scheduling[3][5][6].
