Overview

Dendritic cells + OK-432 is a combination immunotherapy product consisting of autologous or allogeneic dendritic cells (DCs) that have been matured and activated ex vivo using the biological response modifier OK-432 (Picibanil), a lyophilized preparation of penicillin-inactivated Streptococcus pyogenes. This combination is used as an anticancer immunotherapy. The mechanism involves the maturation and activation of DCs by OK-432 through Toll-like receptor 4 (TLR4)-MD2 signaling, leading to enhanced expression of co-stimulatory molecules (CD40, CD80, CD83, CD86), increased production of Th1 cytokines such as IL-12 and IFN-gamma, and improved antigen presentation capacity. These mature DCs are then administered to patients to induce robust tumor-specific cytotoxic T lymphocyte responses. Clinical studies have shown this approach can enhance antitumor immunity in various cancers including hepatocellular carcinoma and colorectal cancer[1][2][3][5].
