Overview

This is a combination cell therapy approach that integrates three distinct immunotherapeutic modalities: - **Dendritic vaccine**: Dendritic cell (DC) vaccines are designed to stimulate the immune system by presenting tumor-associated antigens to T cells, thereby priming and activating cytotoxic lymphocytes against cancer cells. These vaccines can be generated from either autologous or allogeneic sources and are often used in post-remission settings for hematologic malignancies such as acute myeloid leukemia (AML)[1][4][5]. - **Allogeneic hematopoietic stem cells**: Allogeneic hematopoietic stem cell transplantation (allo-HCT) involves transferring stem cells from a healthy donor to the patient, reconstituting the recipient’s immune system and providing a graft-versus-tumor effect. This is commonly used in treating various blood cancers[4][5]. - **Cytotoxic lymphocytes**: Cytotoxic lymphocyte therapies involve infusing T cells or natural killer (NK) cells capable of directly killing malignant or infected target cells. These may be derived from donors or expanded ex vivo. The rationale for combining these modalities is to leverage the antigen-presenting capacity of dendritic vaccines, the immune reconstitution and anti-leukemic effects of allo-HCT, and the direct tumor-killing activity of cytotoxic lymphocytes. Clinical studies have explored combinations such as DC vaccination after allo-HCT[4][5], with some trials reporting induction of specific T-cell responses without increased adverse events.
