Qdenga (TAK-003) is a live-attenuated tetravalent dengue vaccine developed by Takeda. It is constructed using a live-attenuated dengue serotype 2 virus (DENV-2) as the genetic backbone. The vaccine contains attenuated strains for all four dengue virus serotypes (DENV-1, DENV-2, DENV-3, and DENV-4). For serotypes 1, 3, and 4, the pre-membrane (prM) and envelope (E) genes of DENV-2 are replaced with those of the other serotypes, creating chimeric viruses, while the DENV-2 component is the attenuated parent strain itself. Qdenga is designed to elicit both humoral and cellular immune responses against all four serotypes in both dengue-naive and dengue-exposed individuals. It is administered subcutaneously in a two-dose regimen and has been approved in multiple regions, including the European Union, the United Kingdom, Indonesia, and Brazil, for the prevention of dengue disease caused by any of the four serotypes.
