Denikitug (GS-1811, JTX-1811) is an investigational monoclonal antibody that targets C-C chemokine receptor type 8 (CCR8). It is designed to selectively deplete CCR8-expressing regulatory T cells (Tregs) within the tumor microenvironment through mechanisms including direct inhibition of CCR8 and antibody-dependent cell-mediated cytotoxicity (ADCC). By reducing immunosuppressive Tregs in tumors, denikitug aims to enhance anti-tumor immune responses. The drug is being developed for the treatment of advanced solid tumors and is currently in Phase 1 clinical trials as both monotherapy and in combination with zimberelimab, an anti-PD-1 monoclonal antibody. Denikitug was originally developed by Jounce Therapeutics and further developed by Gilead Sciences[1][3][5].
