Denosumab is a fully human monoclonal IgG2 antibody that targets and binds with high affinity and specificity to RANKL (receptor activator of nuclear factor kappa-B ligand), a key mediator in the formation, function, and survival of osteoclasts—the cells responsible for bone resorption. By inhibiting RANKL from activating its receptor RANK on the surface of osteoclasts and their precursors, denosumab reduces bone resorption and increases bone mass. It is used primarily for the treatment of osteoporosis in postmenopausal women and men at high risk for fracture; prevention of skeletal-related events in patients with multiple myeloma or solid tumors metastatic to bone; treatment of giant cell tumor of bone that cannot be surgically removed; management of hypercalcemia due to malignancy refractory to bisphosphonates; as well as prevention or treatment of therapy-induced bone loss (e.g., from corticosteroids or hormone deprivation therapies). Denosumab was developed by Amgen[1][5][6][9].
