Drug intelligence / Profile preview
DENV2 NS1 mRNA-LNP vaccine is an experimental mRNA-based vaccine candidate designed to protect against Dengue virus serotype 2 (DENV2) infection. Developed through a collaboration between the Yale School of Medicine and the University of Pennsylvania, the vaccine is formulated with lipid nanoparticles (LNPs) and encodes the non-structural protein 1 (NS1) of DENV2. Unlike traditional live-attenuated dengue vaccines that target structural proteins and carry a risk of inducing antibody-dependent enhancement (ADE) upon secondary infection, this vaccine induces non-neutralizing anti-NS1 antibodies. These antibodies protect against severe disease and vascular leakage while evading ADE, offering a safer vaccination strategy against lethal dengue challenge.
