Overview

4-Deoxypyridoxine is a synthetic analog of vitamin B6 (pyridoxine) and acts as a vitamin B6 antagonist. Its principal mechanism is inhibition of pyridoxal kinase, an enzyme critical in the regeneration and function of vitamin B6. This antagonism causes vitamin B6 deficiency, which suppresses immune function and produces metabolic derangements. As an immunosuppressant, deoxypyridoxine has demonstrated utility in animal models, such as reducing chronic granuloma formation and dampening cytokine production (TNF-α, IL-1, IL-6). Additionally, it inhibits sphingosine-1-phosphate lyase, proposed to increase the viability of pancreatic islet tissue by reducing apoptosis in transplantation contexts. It is considered toxic, especially during embryonic development, and is not used therapeutically in humans.[1][3][5][9]
