Overview

Deoxyspergualin is a synthetic analogue of the antitumor antibiotic spergualin, originally isolated from *Bacillus laterosporus*. It is classified as an immunosuppressive agent and has been primarily investigated for its ability to prevent and treat organ transplant rejection, as well as for autoimmune diseases. Mechanistically, deoxyspergualin inhibits the proliferation of T cells by blocking their progression from G0/G1 to S and G2/M phases in response to interleukin-2 (IL-2) stimulation. It binds heat shock proteins such as Hsc70 and Hsp90, partially inhibits nuclear transport of NF-kB, reduces cytokine production (including IL-6 and TNF-alpha), decreases antibody production, and impairs early T and B cell development. Deoxyspergualin has shown efficacy in reversing acute graft rejection resistant to other immunosuppressants in clinical trials[1][3][5][8].
