deoxythymidine monophosphate is a nucleotide, specifically the monophosphate ester of the nucleoside thymidine, which itself consists of the nucleobase thymine attached to deoxyribose. Its structure comprises a phosphate group, deoxyribose sugar, and thymine base[1][3][7]. It is one of the four deoxyribonucleotides required for the synthesis of DNA and is a critical fundamental metabolite in cellular processes. Therapeutically, deoxythymidine monophosphate has been investigated as a supplement for genetic mitochondrial diseases caused by thymidine kinase 2 (TK2) deficiency, where it acts by bypassing defective nucleotide salvage pathways, restoring mitochondrial DNA levels, and improving biochemical and phenotypic outcomes in preclinical models[5].
