DEP SN38 is a novel, patented nanoparticle (dendrimer-based) formulation of the active metabolite of irinotecan (SN-38), developed to improve solubility and direct delivery to tumor tissue. By attaching SN-38 to a dendrimer carrier, this drug avoids the metabolic activation and toxic byproducts associated with conventional irinotecan therapy. The mechanism of action is inhibition of topoisomerase I—SN-38 stabilizes the cleavable complex between topoisomerase I and DNA, resulting in DNA breaks and inhibition of DNA replication in cancer cells. Clinical trials have shown promising antitumor activity with improved tolerability compared to standard irinotecan in heavily pre-treated patients with advanced solid tumors such as colorectal cancer (CRC), platinum-resistant high-grade serous ovarian cancer (HGSOC), and breast cancer.
