Depigmented-polymerised grass pollen extract is a chemically modified allergen preparation used for subcutaneous allergen immunotherapy (SCIT) in patients with respiratory allergies such as allergic rhinitis and allergic asthma due to sensitization to grass pollens. The process involves removing pigment molecules and polymerizing the proteins in the pollen extracts, which reduces their IgE-binding capacity while preserving T-cell reactivity. This modification aims to decrease the risk of severe allergic reactions during immunotherapy while maintaining or enhancing clinical efficacy[3][5]. The primary mechanism is immune tolerance induction by repeated exposure to modified allergens, leading to increased production of blocking antibodies (such as IgG4) and reduced sensitivity of effector cells involved in allergy. Depigmented-polymerised extracts are considered safer than native extracts and allow higher doses for improved efficacy[3][5]. These preparations are typically administered via subcutaneous injection over several years.
