Overview
DepoCyte is a sustained-release liposomal formulation of the antimetabolite chemotherapy agent cytarabine, specifically engineered for intrathecal administration. Utilizing the proprietary DepoFoam technology, the drug encapsulates cytarabine in multivesicular liposomes, which allows for a slow, controlled release into the cerebrospinal fluid (CSF). This formulation significantly extends the half-life of cytarabine in the CNS, maintaining therapeutic concentrations for up to 14 days and reducing the required frequency of lumbar punctures compared to standard cytarabine. Cytarabine itself is a pyrimidine analog that, once phosphorylated, inhibits DNA polymerase and incorporates into DNA, thereby disrupting DNA synthesis and repair in rapidly dividing cancer cells. DepoCyte is primarily indicated for the treatment of lymphomatous meningitis and has been investigated by the PETHEMA Foundation for its efficacy in treating central nervous system (CNS) relapse in patients with acute lymphoblastic leukemia (ALL) and aggressive non-Hodgkin lymphomas, such as Burkitt lymphoma.
