Depulfavirine is a small molecule non-nucleoside reverse transcriptase inhibitor (NNRTI) developed for the treatment of HIV-1 infection. It is also known by code names such as RO-0335 and VM 1500A, and is marketed under the brand name Elpida in some regions. Depulfavirine acts by selectively inhibiting HIV reverse transcriptase, thereby preventing viral replication. The drug demonstrates potent activity against wild-type HIV as well as many NNRTI-resistant strains, with a high genetic barrier to resistance development[1][3][4]. Depulfavirine has been formulated for both oral and long-acting injectable administration; the latter allows for once-monthly or less frequent dosing in combination antiretroviral therapy[4][5]. Clinical studies have shown that regimens based on depulfavirine are at least as effective as efavirenz-based therapies but with improved tolerability and safety profiles[4]. The drug was originally developed by Roche and further advanced by Viriom.
