Overview

Der p 2 is a major allergen protein derived from the house dust mite Dermatophagoides pteronyssinus. It belongs to the NPC2 family and contains an MD-2-related lipid recognition (ML) domain. The protein has a molecular weight of approximately 14–15 kDa and is found in high concentrations in the digestive tract and fecal pellets of D. pteronyssinus. Structurally, it is a single-domain β-sandwich protein stabilized by three disulfide bonds and features a hydrophobic cavity that binds aliphatic chains. Der p 2 mimics human MD-2 (a co-receptor for Toll-like receptor 4), allowing it to interact with the innate immune system and contribute to allergic sensitization. Exposure to Der p 2 is highly predictive of immediate asthmatic responses in sensitized individuals; IgE reactivity rates among house dust mite-allergic patients are very high (upwards of ~80–100%). It exhibits strong cross-reactivity with other group 2 allergens such as Der f 2 from Dermatophagoides farinae[3][5][2]. Recombinant or purified natural forms are used for research, diagnostics, and as components in allergen-specific immunotherapy.
