DerG-PG275Cit is a preclinical therapeutic vaccine candidate developed by CEL-SCI for the treatment of rheumatoid arthritis. It is a fusion protein and part of the LEAPS (Ligand Epitope Antigen Presentation System) platform, designed to modulate immune responses by targeting CD4 T cells. The vaccine incorporates a citrullinated epitope from cartilage proteoglycan (PG), which is relevant in autoimmune pathogenesis. In animal models of rheumatoid arthritis, DerG-PG275Cit has demonstrated immunomodulatory effects—specifically upregulating regulatory T cells and anti-inflammatory cytokines such as IL-4 and IL-10—without eliciting significant antibody responses. Its mechanism involves shifting pro-inflammatory Th1/Th17 responses toward an anti-inflammatory/regulatory profile via CD4 modulation[1][2][6][7][8]. The drug remains in preclinical development.
