Dermatophagoides pteronyssinus allergoid is a subcutaneous allergen immunotherapy (SCIT) product developed by Allergopharma GmbH & Co. KG. It is designed for the treatment of IgE-mediated allergic diseases caused by the house dust mite *Dermatophagoides pteronyssinus*, including allergic bronchial asthma, allergic rhinitis, and allergic rhinoconjunctivitis. The product contains a purified extract of *D. pteronyssinus* that has been chemically modified (polymerized) using glutaraldehyde to form an "allergoid." This modification significantly reduces the allergen's IgE-binding capacity, thereby decreasing the risk of systemic allergic reactions during administration, while maintaining the T-cell epitopes necessary to induce immunological tolerance. The allergoid is adsorbed onto aluminium hydroxide, which acts as a depot adjuvant to enhance the immune response. The mechanism of action involves shifting the immune system from a Th2-dominated allergic response toward a Th1 and regulatory T-cell (Treg) profile, leading to the production of allergen-specific IgG4 antibodies and long-term desensitization.
