Dermatophagoides pteronyssinus allergoid (glutaraldehyde-modified) is a modified allergen extract used in allergen immunotherapy (AIT) for the treatment of house dust mite-induced allergic rhinitis and rhinoconjunctivitis. Developed by ROXALL Medizin GmbH, this allergoid is produced by treating the native allergens of the European house dust mite with glutaraldehyde. This chemical modification induces polymerization, which significantly reduces the allergen's IgE-binding capacity (allergenicity) while preserving its ability to stimulate T-cells (immunogenicity). By reducing IgE-mediated side effects, the allergoid allows for the administration of higher allergen doses, facilitating a more rapid and effective induction of immunological tolerance. The treatment works by shifting the immune response from a Th2-dominated allergic profile to a Th1 and regulatory T-cell (Treg) profile, characterized by the production of blocking IgG4 antibodies and anti-inflammatory cytokines like IL-10. It is typically administered as a subcutaneous injection (SCIT) and is often adsorbed onto aluminum hydroxide to enhance its immunogenic effect.
