Overview

Dersalazine sodium is a locally acting small molecule drug developed as a novel anti-inflammatory agent for the treatment of ulcerative colitis. It is a potent antagonist of platelet activating factor (PAF) and also inhibits the production of proinflammatory cytokines such as IL-1β and TNF-α. Dersalazine sodium is an azo-bonded compound that combines a PAF antagonist (UR-12715) with 5-aminosalicylic acid (5-ASA). After oral administration, it remains largely unabsorbed until reaching the large intestine, where bacterial reduction releases its active components. In preclinical models and early clinical trials, it demonstrated downregulation of inflammatory mediators including IL-1β, IL-6, and IL-17 in colonic tissue. Phase I/II studies in patients with ulcerative colitis showed promising efficacy signals and no serious adverse reactions; however, development has not progressed recently[2][4][5][8].
