Cincera Therapeutics is developing an oral small-molecule inhibitor of Dihydroceramide desaturase 1 (Des-1), an enzyme that catalyzes the final step in the de novo synthesis of ceramides. By inhibiting Des-1, the drug aims to modulate the levels of bioactive lipids, specifically reducing ceramides and increasing dihydroceramides, to mitigate lipotoxicity, inflammation, and fibrosis. This therapeutic approach is being targeted at metabolic and fibrotic conditions, including non-alcoholic steatohepatitis (NASH) and fibrosis of the liver, kidney, lung, and heart. The program is currently in preclinical development, supported by funding from the Australian government's Biomedical Translation Bridge Program and Brandon Capital's Medical Research Commercialisation Fund.
