Desaminotyrosine (DAT) is a microbial metabolite produced by the gut microbiota, specifically through the degradation of dietary flavonoids by species such as *Flavonifractor plautii*. It acts as a potent immunomodulatory and tissue-protective agent, primarily by enhancing type I interferon (IFN-I) signaling through the interferon-alpha/beta receptor (IFNAR). DAT has demonstrated therapeutic potential in preclinical models for a variety of conditions, including viral infections (such as Influenza and Respiratory Syncytial Virus), obesity, graft-versus-host disease (GvHD), and cancer immunotherapy, where it enhances the efficacy of immune checkpoint inhibitors. Mechanistically, DAT also promotes intestinal regeneration via the mTORC1 and STING pathways and exhibits antioxidant properties by modulating the cellular NADP+/NADPH ratio, which contributes to its cardioprotective effects against myocardial ischemia/reperfusion injury.
