Descartes-15 is a next-generation autologous B cell maturation antigen (BCMA)-directed mRNA-engineered chimeric antigen receptor T-cell therapy (CAR-T) developed by Cartesian Therapeutics. It is designed to have predictable and controllable pharmacokinetics with technological advances that enhance CAR stability even in the presence of target-driven suppression of CAR. Unlike conventional DNA-based CAR-T therapies, Descartes-15 does not require preconditioning chemotherapy and avoids the risk of genomic integration by using mRNA engineering rather than integrating vectors. Preclinical studies have shown that Descartes-15 achieves approximately ten-fold higher CAR expression and selective target-specific killing compared to earlier candidates such as Descartes-08. The drug is currently being evaluated in Phase 1 clinical trials for relapsed/refractory multiple myeloma and has preclinical development for autoimmune disorders and solid tumors[1][2][3][5][6].
