desHis1Pro4Glu9-glucagon is a synthetic peptide derived from native glucagon through specific amino acid modifications (desHis1, Pro4, Glu9). It functions as a potent and specific antagonist of the glucagon receptor (GCGR). By blocking the GCGR, it inhibits the downstream signaling pathway, including the activation of adenylyl cyclase and the subsequent increase in cyclic AMP (cAMP), which normally leads to increased hepatic glucose production. Preclinical studies in animal models of diabetes have demonstrated its ability to lower circulating blood glucose and glucagon levels, improve glucose tolerance, and elevate plasma insulin. It has also been observed to positively influence alpha-cell turnover and lineage. This peptide is being investigated for its therapeutic potential in treating diabetes, particularly obesity-driven forms, and related metabolic disorders.
