Overview

Desipramine + fluoxetine is a combination of two antidepressant medications used together, primarily studied for the treatment of major depressive disorder. Desipramine is a tricyclic antidepressant (TCA) that acts mainly by inhibiting the reuptake of norepinephrine, thereby increasing its activity in the brain. Fluoxetine is a selective serotonin reuptake inhibitor (SSRI) that increases serotonin levels by blocking its reabsorption into neurons. The combination has been investigated for potential synergistic effects, with some studies suggesting it may lead to more rapid and effective symptom improvement compared to monotherapy with either agent alone. However, combining these drugs can increase the risk of side effects and drug interactions due to pharmacokinetic interactions—fluoxetine inhibits CYP2D6, which can raise desipramine plasma levels and increase toxicity risk[1][2][5]. This combination should be used under close medical supervision.
