Drug intelligence / Profile preview
Desmethyl pateamine A (DMPatA, MZ735) is a novel, potent small molecule inhibitor of the eukaryotic translation initiation factor 4A (eIF4A) RNA helicase. Derived as a synthetic analog of the marine sponge-derived natural product pateamine A, DMPatA acts as an interfacial inhibitor that clamps eIF4A onto RNA, thereby inhibiting cap-dependent translation initiation. This translation inhibition leads to the depletion of key short-lived oncoproteins, such as c-MYC and cyclin D1. DMPatA has demonstrated significant synergistic antitumor activity when combined with class I histone deacetylase (HDAC) inhibitors, such as romidepsin, in preclinical models of pancreatic ductal adenocarcinoma (PDAC). This combination prolongs histone hyperacetylation, induces replication stress, and triggers DNA damage, leading to pancreatic cancer cell death.
