Overview

PI-0824 is a synthetic peptide-based immunotherapy developed for the treatment of pemphigus vulgaris (PV), a rare autoimmune blistering skin disease. The drug is designed to reduce the production of pathogenic antibodies by specifically targeting disease-causing components of the immune system, aiming to induce anergy in disease-associated T-cells. Its mechanism involves modulation of immune responses against desmoglein 3 (DSG3), a key autoantigen in PV. PI-0824 was intended as an alternative to high-dose steroids and broad immunosuppressants, with hopes to minimize side effects associated with current therapies. The therapy was granted orphan drug designation for PV but development has been discontinued after early-phase clinical trials[1][2][5][7].
